Um, what do you like to do? My recommendation, sans further information, is: WALK. Get a decent walking guidebook and an A-Z, and wear out your shoe leather.
The single most awesome place in the entire city is The Black Friar, the most beautiful pub in the world, an Art Deco masterpiece just at the north end of Blackfriars Bridge.
Portobello Road Market (flea market). Leadenhall Market in the City (lovely arcade, upscale). Smithfield Market (the main meat market; get there at 6 AM to see the action). A curry on Brick Lane. Borough Market in Southwark (food). The George Inn in Southwark. The Lamb and Flag in Covent Garden. The Prospect of Whitby in Wapping. Ye Olde Cheshire Cheese (I like pubs). Six Dials and Neal's Yard. Pellici's Cafe on Bethnal Green Road (http://everything2.com/index.pl?node_id=834955) (I hope it's still there; the New Piccadilly is sadly gone).
Stick your head in every single one of Christopher Wren's churches in London (http://en.wikipedia.org/wiki/List_of_Christopher_Wren_churches_in_London). At St. Paul's climb the stairs inside the domes.
Explore the narrow pedestrian lanes in the City, like Gough Square (home of Dr. Johnson) off of Fleet Street.
John Soames Museum. Tate Modern. British Museum. Victoria and Albert Museum. The Geffrye Museum in Shoreditch. Kew Gardens. Skip the Tower of London; it's tacky (especially the crown jewels). Also, skip the Changing of the Guard. Go shopping on Oxford Street instead.
In summer, go to the cricket. In other seasons, go to the football; tickets to the top clubs are hard to come by but you should be able to get into Charlton or West Ham or Millwall or Crystal Palace.
